ScientificBackground

  • MostoftheeffectsofcAMParemediatedthroughthephosphorylationoftargetproteinsonserineorthreonineresiduesbythecAMP-dependentproteinkinase(AMPK).TheinactiveholoenzymeofAMPKisatetramercomposedoftworegulatoryandtwocatalyticsubunits.ThemammaliancatalyticsubunithasbeenshowntoconsistofthreePKAgeneproducts:C-alpha,C-beta,andC-gamma.TwoPKAisoformsexist,designatedtypesIandII,whichdifferintheirdimericregulatorysubunits,designatedRIandRII,respectively.FurThermore,thereareatleastfourdifferentregulatorysubunits:RI-alpha,RI-beta,RII-alpha,andRII-beta.ThecAMPcausesthedissociationoftheinactiveholoenzymeintoadimerofregulatorysubunitsboundtofourcAMPandtwofreemonomericcatalyticsubunits.ThecatalyticsubunitC-betaofPKA(PKAcb)isamemberoftheSer/ThrproteinkinasefamilyandisacatalyticsubunitC-betaofAMPK.Berubeetal.assignedthePKAcbtohumanchromosome1bySouthernblotanalysisofsomaticc
