ScientificBackground

  • ProteinkinaseCdelta(PKCdelta)isamemberoftheproteinkinaseC(PKC)familyofserine-threoninekinases.Itisa79kdproteinkinasethatshowsstrictdependenceonthepresenceofphospholipids,butshowsnoactivationbyCa2+(1).PhosphatidylinositolisthemostpotentactivatorofPKCdelta.Apparentkineticconstantsforsyntheticoligopeptides(MBP4-14,EGFRpeptideandepsilon-peptide)arequitedifferentbetweenPKCdeltaandotherPKCs.NorthernblotanalysisindicatedthatPKCdeltaiswidelydistributedinalmostallthetissuesandisamajorisoformofPKCexpressedinhemopoieticcells(2).PKCdeltaisinvolvedinfundamentalcellularfunctionsregulatedbydiacylglycerolsandmimickedbyphorbolesters.PKCdeltaispartiallyassociatedwiththeinsolublefractionincellsevenintheabsenceofphorbol12-myristate13-acetate(PMA).UponPMAstimulation,bothittranslocatetotheinsolublefractionofcellhomogenates(3).OverexpressionofPKC-deltainducessignificantchanges
