ProductBackground:Autotaxin,alsoknownasATX,ENPP2,lysophospholipaseD(lysoPLD),phosphodiesterase1αandplasmacellglycoprotein-1,isasecretedglycoproteinthatiswidelyexpressedwithhighlevelsintheserum.ViaitslysoPLDactivity,autotaxinhydrolyzeslysophosphatidylcholine(LPC)togeneratethephospholipidgrowthfactorlysophosphatidicacid(LPA).Theenzyme’ssameactivityhydrolyzessphingosylphosphorylcholine(SPC)toformsphinosine-1-phosphate(S1P).Autotaxinwasfirstisolatedastheautocrinemotilityfactorsecretedfrommelanomacells.Atthattime,theenzyme’slysoPLDactivityhadnotbeenidentified,soitwasclassifiedbyhomologytotheecto-nucleotidepyrophosphatase/phosphodiesterase(NPP)familyofenzymeswhosemembershydrolyzephosphodiesterbondsinvariousnucleotidesandnucleotidederivatives.Itwasinitiallyunclearhownucleotidehydrolysiscouldleadtothestimulationofcellmotility.ThismysterywassolvedwhenautotaxinwasdiscoveredtobeidenticaltoserumlysoPLD.Sincethen,thecancer-relatedactivitiesofautotaxin,atleastinculturedcells,havebeenattributedtotheenzyme’slysoPLDactivity.Inadditiontocancer,autotaxinhasbeenimplicatedinanumberofdiseasesincludingobesity,arthritis,multiplesclerosis,Alzheimer’sdiseaseandneuropathicpain.
